These scientific breakthroughs recently published in the prestigious&nbsp;<em>PLoS Pathogens<\/em>&nbsp;journal represent two important clues to understanding the cellular and molecular mechanisms governing the parasitic infections that cause leishmaniasis, a neglected tropical disease endemic in one hundred countries.<\/strong><\/p>\n\n\n\n<div class=\"wp-block-image\"><figure class=\"aligncenter size-large is-resized\"><img loading=\"lazy\" decoding=\"async\" src=\"https:\/\/dev.inrs.ca\/wp-content\/uploads\/2020\/11\/NEWS_INRS-professors-team-unveils-new-Leishmania-virulence-strategies_01.jpg\" alt=\"\" width=\"690\" height=\"194\"\/><\/figure><\/div>\n\n\n\n<p><em>Leishmania<\/em>&nbsp;uses macrophages, a type of white blood cell that neutralizes pathogenic micro-organisms, as host cells to circumvent the immune system. Upon infecting the macrophage, Leishmania enters a vacuole that it hijacks by manipulating the host cell membrane fusion machinery. Research results revealed that GP63, a zinc-dependent metalloprotease found on the surface of Leishmania, plays a central role in this process.&nbsp;<\/p>\n\n\n\n<p>By promoting the cleavage of the VAMP8 membrane fusion regulator, GP63 allows&nbsp;<em>Leishmania<\/em>&nbsp;major to evade LC3-associated phagocytosis, a process whose purpose is to boost the antimicrobial properties of macrophages. &nbsp;Researchers have also found that cysteine peptidase CPB, which the parasite needs to survive in the macrophage, controls the virulence of&nbsp;<em>Leishmania<\/em>&nbsp;mexicana by regulating GP63 expression through an as yet unknown mechanism. &nbsp;<\/p>\n\n\n\n<p>These two proteases occupy a central role in the biology of&nbsp;<em>Leishmania<\/em>&nbsp;and contribute to the formation and growth of parasitophorous vacuoles where the parasite develops and replicates. Researchers have thus uncovered a new&nbsp;<em>Leishmania<\/em>&nbsp;virulence mechanism heavily influenced by GP63 and CPB proteases. <\/p>\n\n\n\n<div class=\"wp-block-media-text alignwide is-stacked-on-mobile\"><figure class=\"wp-block-media-text__media\"><img decoding=\"async\" src=\"https:\/\/dev.inrs.ca\/wp-content\/uploads\/2020\/11\/NEWS_INRS-professors-team-unveils-new-Leishmania-virulence-strategies_02.jpg\" alt=\"\"\/><\/figure><div class=\"wp-block-media-text__content\">\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\"><p>\u201cOur results give us a better understanding of&nbsp;<em>Leishmania<\/em>&nbsp;pathogenesis and identify a new strategy used by intracellular pathogens to disrupt the host cell\u2019s antimicrobial.\u201d<\/p><cite>Professor Descoteaux<\/cite><\/blockquote>\n<\/div><\/div>\n\n\n\n<p>This discovery will be useful in developing preventive or therapeutic measures against leishmaniasis.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><strong>Two articles have been published in&nbsp;<a href=\"http:\/\/journals.plos.org\/plospathogens\/\" target=\"_blank\" rel=\"noreferrer noopener\"><em>PLoS Pathogens<\/em><\/a>:<\/strong><\/h3>\n\n\n\n<p>\u201c<a href=\"http:\/\/journals.plos.org\/plospathogens\/article?id=10.1371\/journal.ppat.1005658\" target=\"_blank\" rel=\"noreferrer noopener\">Cysteine Peptidase B Regulates&nbsp;<em>Leishmania mexicana<\/em>&nbsp;Virulence through the Modulation of GP63 Expression<\/a>\u201d presents the results of research carried out by Pierre-Andr\u00e9 Casgrain (master\u2019s student), Caroline Martel, W. Robert McMaster, Jeremy C. Mottram, Martin Olivier, and Albert Descoteaux. The researchers received financial support from the Canadian Institutes of Health Research, the Medical Research Council, and the Centre for Host-Parasite Interactions.&nbsp;<a href=\"http:\/\/dx.doi.org\/10.1371\/journal.ppat.1005658\" target=\"_blank\" rel=\"noreferrer noopener\">doi:10.1371\/journal.ppat.1005658<\/a><\/p>\n\n\n\n<p>\u201c<a href=\"http:\/\/journals.plos.org\/plospathogens\/article?id=10.1371\/journal.ppat.1005690#abstract1\" target=\"_blank\" rel=\"noreferrer noopener\"><em>Leishmania<\/em>&nbsp;<em>major&nbsp;<\/em>Promastigotes Evade LC3-Associated Phagocytosis through the Action of GP63<\/a>\u201d was written by Christine Matte (research assistant), Pierre-Andr\u00e9 Casgrain (master\u2019s student), Olivier S\u00e9guin (doctoral student), Neda Moradin (doctoral student), Wan Jin Hong, and Albert Descoteaux.
